两株产紫杉醇内生真菌A11和J23
的初步形态学鉴定
摘要
紫杉醇是从红豆杉科植物红豆杉的干燥根,枝叶以及树皮中提取的物质,对于卵巢癌和乳腺癌,肺癌、大肠癌、黑色素瘤、头颈部癌、淋巴瘤、脑瘤等疾病都有一定疗效。由于紫杉醇的来源主要是从红杉树中提取,因此造成了红豆杉树遭到毁灭性的砍伐。为了保护红豆杉树和解决紫杉醇的来源问题,对已分离的产紫杉醇内生真菌进行形态鉴定,以便进行产紫杉醇内生菌的选育。首先进行选株,选择无污染、菌落长势较好的菌种作为接种菌种。其次,在无菌条件下,把选好的菌株插入事先配置好的马铃薯葡萄糖琼脂培养基上。然后,把培养基置于恒温箱(24℃)中培养,并定期观察菌落生长情况,等菌落长到一定时期根据其特征,并在光学显微镜下观察菌丝形态,是否有孢子生成及其形态特征。查阅《真菌鉴定手册》进行初步鉴定。对两个菌株的初步形态鉴定结果是:菌种A11可能属于半知菌亚门串珠霉属;J23可能为头孢霉属。

TWO STRAINS OF ENDOPHYTIC FUNGI OF PRODUCING TAXOL AND J23 A11
THE PRELIMINARY MORPHOLOGICAL IDENTIFICATION
ABSTRACT
Paclitaxel from yew families of dry fields of plant roots, leaves and bark extract substances, for ovarian and breast cancer, lung cancer, colorectal cancer, head and neck cancer, melanoma, lymphoma, brain tumors and other diseases have certain curative effect.Because of the main sources of taxol is extracted from the redwood, so caused the ormosia fir trees were devastating logging.In order to protect the ormosia fir and solve the problem of sources of taxol, for the separation of morphological identification of endophytic fungi of producing taxol, selective breeding to produce taxol endogenous bacteria.First to choose plants, choose non-polluting, colony growth better strains as inoculated strains. Secondly, under aseptic conditions, inserted the selected strains previously configured on potato dextrose AGAR medium.Then put culture medium in the constant temperature box (24 ℃), and regularly observed colony growth, such as to a certain period according to its characteristics of colony, and observed under optical microscope mycelial morphology, spore formation and its morphological characteristics.Refer to handbook and fungal identification to carry on the preliminary appraisal. Preliminary form of the two strains identification result: strains of A11 may be imperfect fungi door beaded mildew genera; J23 for cephalosporin mildew genera.
